چکیده

We present an effective model All-Trans Retinoic Acid (ATRA)-induced differentiation of HL-60 cells. The model describes a key architectural feature of ATRA-induced differentiation, positive feedback between an ATRA-inducible signalsome complex involving many proteins including Vav1, a guanine nucleotide exchange factor, and the activation of the mitogen activated protein kinase (MAPK) cascade. The model, which was developed by integrating logical rules with kinetic modeling, was significantly smaller than previous models. However, despite its simplicity, it captured key features of ATRA induced differentiation of HL-60 cells. We identified an ensemble of effective model parameters using measurements taken from ATRAinduced HL-60 cells. Using these parameters, model analysis predicted that MAPK activation was bistable as a function of ATRA exposure. Conformational experiments supported ATRAinduced bistability. These findings, combined with other literature evidence, suggest that positive feedback is central to a diversity of cell fate programs.

اثر هم افزایی کاربرد توأم زهر زنبور عسل و25،1- دی هیدروکسی ویتامینD3 برالقای تمایز رده ی سلولی سرطانی پرومیلوسیتی HL-60

Introduction & Objective: Acute promyelocytic leukemia (APL) is a kind of acute leukemia characterized by a balanced t (15, 17) translocation which fails to develop into mature cells and proliferate in an unregulated fashion. In the recent years, in addition to combinatoral chemotherapy to treat unmature cancerous cells, differentiation therapy by differentiating agents as a novel procedure ...

اثر زهر زنبور عسل برروی قدرت تمایزی-D آلفا توکوفرول سوکسینات (ویتامین E) در رده سلول سرطانی حاد پرومیلوسیت HL-60

Background: Acute promyelocytic leukemia is the most malignant type of myeloid leukemia characterized by chromosomal translocation (15 and 17) and also blocking the cells in promyelocytic stage of differentiation into myeloid. Nowadays, differentiation therapy is used to treat leukemia. Previous studies indicate that vitamin E inhibits proliferation and also induces differentiation of HL-60 cel...

Gnidilatimonoein from Daphne mucronata induced differentiation and apoptosis in leukemia cell lines

Among various treatments, plants play a crucial role in cancer chemotherapy. Base on literature data, different species of the thymeleaeceae family have been found to be good sources for anticancer agents. Therefore, our laboratory has initiated a research project on evaluating the anticancer properties of the Iranian medical plant, Daphne mucronata (Thymeleaeceae). Gnidilatimonoein is a new di...

Background and purpose: Cancer is one of the leading causes of mortality worldwide. Acute promyelocytic leukemia, which is the most common type of blood cancer in adults, is the most prevalent type of acute leukemia. This type of cancer is characterized by uncontrolled proliferation, inhibited differentiation of immature myloied progenitors (promyelocytes), and accumulation of abnormal promye...
